Oxetanocin-G (OXT-G), a potent antiviral agent, is a novel nucleoside isolated from the culture filtrate of Bacillus megaterium. We investigated the antiviral effect of oral administration of OXT-G for five days on woodchuck hepatitis virus (WHV), in vivo, using 12 woodchucks. Woodchucks were randomized into each of four treatment groups according to the dose of OXT-G. Two out of six woodchucks treated with 1.0 or 2.0 mg/kg/day of OXT-G died. After treatment with OXT-G, serum levels of WHV-DNA significantly decreased in all woodchucks. However, the antiviral effect was only partial and levels of serum WHV-DNA returned after the cessation of treatment. The amount of viral replicative intermediates was decreased in livers of woodchucks treated with OXT-G. Although further study of the toxicity of this compound would be essential before studies in man can be carried out, OXT-G has potent antiviral activity against WHV and may deserve evaluation as an antiviral agent in the treatment of chronic hepatitis B infections in humans.

The paper deals with the question to what extent increasing tourist and leisure activities are a serious problem for wildlife. It presents the project ''Tourismus und Wild'' carried out between 1990 and 1993. The aim of the project is to measure the effects of different forms of tourism and leisure activities (hiking, paragliding etc.) as a basis for optimizing the relationship between tourism/recreation and wildlife. The studies were carried out on chamois, ibex, alpine marmots and ptarmigan in several areas of the Swiss alps differing in their amount of tourist and leisure activities: a) in the same area in the course of the day and on different days; b) in different sections of an area and in different areas. Several examples from the current study are used for illustration. Hikers and alpine marmots: the example shows that hiking can have a stronger effect than would be expected from the greatly reduced flight distances of the animals. It shows as well that it matters whether hikers keep to the path and whether they are accompanied by a dog or not. Hikers and chamois: moderate hiking activity on a path leading across a preferred grazing area of male chamois influences their use of the area in the course of the day. When hikers are present the chamois avoid the attractive grazing areas. Hikers and ptarmigan: By measuring heart-rate it was shown that bradycardia (decrease in frequency) occurs when a person is approaching the nest. Bradycardia is a subtle indicator for stimuli to which incubating hends in the context of predator avoidance. Paragliders and chamois: in areas which so far have been rarely used by paragliders experimental flights provoked strong reactions (escape to the woods at great distances). The development of the responses is followed up. It is not yet possible to present definite results on the behaviour of animals in areas where paragliding has been carried out for a long time. Criteria are listed to clarify the question as to when certain events have to be considered as a ''serious problem'': reduced survival and/or reproduction rate (even if a direct proof is lacking, as long as there is a strong indication from the analysis of the behaviour); habitat loss; effects on further elements of the habitat (e.g. damage to the vegetation due to overgrazing). There is an urgent need to solve the problem of ''flying objects (particularly paragliders) and wildlife''. In collaboration with the federal office for the environment and the groups concerned (e.g. paragliding association) it is planned to work out measures to limit the effect on wildlife.

Calendar date of the beginning of the growing season at high altitude in the Colorado Rocky Mountains is variable but has not changed significantly over the past 25 years. This result differs from growing evidence from low altitudes that climate change is resulting in a longer growing season, earlier migrations, and earlier reproduction in a variety of taxa. At our study site, the beginning of the growing season is controlled by melting of the previous winter's snowpack. Despite a trend for warmer spring temperatures the average date of snowmelt has not changed, perhaps because of the trend for increased winter precipitation. This disjunction between phenology at low and high altitudes may create problems for species, such as many birds, that migrate over altitudinal gradients. We present data indicating that this already may be true for American robins, which are arriving 14 days earlier than they did in 1981; the interval between arrival date and the first date of bare ground has grown by 18 days. We also report evidence for an effect of climate change on hibernation behavior; yellow-bellied marmots are emerging 38 days earlier than 23 years ago, apparently in response to warmer spring air temperatures. Migrants and hibernators may experience problems as a consequence of these changes in phenology, which may be exacerbated if climate models are correct in their predictions of increased winter snowfall in our study area. The trends we report for earlier formation of permanent snowpack and for a longer period of snow cover also have implications for hibernating species.

Liver DNA specimens from woodchucks kept in captivity, 10 naturally infected with hepatitis virus (WHV) and five WHV-free, were examined for the presence of carcinogen-DNA adducts by 32P-postlabeling. The number of adducts was significantly higher in WHV carriers than in uninfected animals, and the total amounts of adducts per 10(9) nucleotides were also considerably enhanced by WHV infection, when using both butanol extraction (22.2 +/- 7.1 vs. 12.6 +/- 2.8, means +/- S.D.) and nuclease P1 enrichment (8.5 +/- 5.9 vs. 2.8 +/- 1.7). Two individual adducts were also significantly higher in WHV carriers. No significant variation occurred as related to age, sex or time length of captivity. These findings are consistent with our previous studies supporting an enhanced metabolism of chemical hepatocarcinogens in both human and woodchuck hepadnavirus infections. Several significant and remarkable correlations were pointed out by relating DNA adduct data to more than 30 virological, histopathological and metabolic parameters which had been previously evaluated in the same animals. For instance, numbers and/or levels of adducts were positively related to the amounts of virus present in hepatocytes, to cell damage (gamma-glutamyltranspeptidase activity), to the severity of the liver histopathological picture, and to monooxygenase activities, while they were inversely related to cellular glutathione concentrations and to detoxification of the direct-acting mutagen 4-nitroquinoline 1-oxide. The major adduct significantly correlated with the metabolic activation of the aromatic amine 2-aminofluorene and of the heterocyclic amines 3-amino-1-methyl-5H-pyrido(4,3)indole (Trp-P-2) and 2-amino-3,4-dimethylimidazo(4,5-f)quinoline (MeIQ), whereas another adduct significantly correlated with the metabolic activation of the mycotoxin aflatoxin B1. Thus, the enhanced metabolism of chemical hepatocarcinogens and the increased formation of carcinogen-DNA adducts in the liver of WHV carriers appear to represent one of the mechanisms contributing to the association between chronic hepadnavirus infection and development of primary hepatocellular carcinoma.
